In an interview with IGN, Rob Nelson, co-studio head at Rockstar North, explained that Jason and Lucia will always have each other's backs during the story, but nurturing the relationship will be up to the player.
"That's up to the player, and you won't be penalized if you're not interested in the sappy stuff," Nelson said. This basically tells us we can't make them break up, but we can alter their relationship's depth.

Players can also respond to texts while using one character to fortify their bond.
How Does Jason and Lucia's Relationship Affect Gameplay?
Although their relationship won't affect the story, since it is not a requirement, it appears we will see some gameplay changes because of it.
According to an article in The New York Times, if the couple's relationship is strong enough, we will see them holding hands when they are together, such as when leaving a clothing store. We assume other gameplay aspects will show their relationship status, such as other mannerisms, but we can only speculate for the time being.
